After the game is initialized, only these callbacks are called on game lifecycle:

  1. PollAllControlsState
  2. UpdateVideo
  3. MakeRunningGameCalls

This is not my experience. I have always received audio callbacks. Is it possible that your game does not produce audio? Which game specifically? Have you made any local modifications to libmame? The version of mame supported by the latest release of libmame is pretty old, are you possibly using ROMs that are not matched with that version of mame?
